A kind of antimicrobial peptide from Odorrana jingdongensis,preparation and application

Pathogen resistance against antibiotic is increasing rapidly. Antimicrobial peptides have following features: small molecular weight, water-soluble, low antigenicity, temperature stability, particularly with character that microbe difficulty to resistance against them. Antimicrobial peptides may become new generation anti-infective drugs and thus are attracted widespread attention.

Researchers in Chengdu Institute of Biology isolated and purified a novel antimicrobial peptide from skin of Odorrana jingdongensis by using multi-step column chromatographies. Its sequence, NH2-FLPAVLRVAAKVVPTVFCLISKKC –COOH, is revealed by employing tandem mass spectrometry de novo sequencing with the help of Edman degradation. This antimicrobial peptide is a single chain polypeptide with 24 amino acids, of which theoretical monoisotopic molecular mass is 2600.5183 Da ([M+H]+) .

This antimicrobial peptide has good ability against a variety of bacteria, especially to common clinical drug-resistant bacteria such as Methicillin-resistant coagulase negative Staphylococcus, Methicillin-resistant Staphylococcus aureus, Extended-spectrum β-lactamases Escherichia coli, Inhibitor-resistant TEM β-lactamase Escherichia coli and AmpC-β-lactamase-producing Enterobacter cloacae; and also towild type bacteria such as Escherichia coli and Staphylococcus aureus. This antimicrobial peptide can be used to develop antimicrobial drug or antibacterial agent.

"One kind of antimicrobial peptide from Odorrana jingdongensis,preparation and application" was licensed in May 2013,with the Chinese invention patent No. "ZL 201110108702.8".
